This project focused on designing and synthesising hydrolytically stable carbocyclic analogues of Decitabine, mainly as prodrug derivatives to enhance cellular uptake and bioavailability. It involved nucleoside modification, the construction of carbocyclic scaffolds for stability, and the design of prodrugs to improve membrane permeability and intracellular delivery to investigate DNA methyltransferase inhibition. The synthesis included protection-deprotection strategies and nucleoside functionalisation.
Final compounds were purified using HPLC and characterised through ¹H and ¹³C NMR, ³¹P NMR, FTIR, and LC-MS to confirm structure and purity. The prodrug analogues were tested as epigenetic probes and potential anticancer agents.
