Development of a pipeline for drug designing
Identification of druggable target using subtractive and comparative genomics approach which majorly includes essentiality analysis, virulence analysis, network analysis, host-pathogen interaction analysis, druggability of target.
Designing of genome scale metabolic model of pathogen for in silico gene knockout and metabolomic analysis.
Design of potential drug or small molecule inhibitor against the proposed druggable targets via active site prediction, docking (schrodinger), molecular dynamic simulation, energy profile analysis, principle component analysis (PCA).
In vitro validation of proposed drug through cell culture experiments such as growth analysis (growth curve, disc difussion) and virulence analysis (hydrolytice enzyme assay, biofilm formation assay, adhesion assay)
