MSc Dissertation – Isolation of Bacterial Endophytes from Halophytic Plant Suaeda sp. at Department of Biotechnology, Kurukshetra University (2026-01 – 2026-06)

- Isolated 44 morphologically distinct endophytic bacteria from root (16), stem (15) and leaf (13) tissues using plant-part inoculation and maceration methods on nutrient agar.
- Performed surface sterilization with sterility checks, aseptic culturing under laminar air flow, and purified isolates by repeated streaking; maintained pure cultures on agar slants.
- Characterized isolates by colony morphology and Gram staining (majority Gram-negative rods) and by catalase, oxidase, citrate, urease, starch hydrolysis and methyl red tests.
- Assessed salt tolerance on nutrient agar with 0–15% NaCl; isolates grew up to 10% NaCl, identifying halotolerant strains.
- Recorded, tabulated and analyzed experimental observations.
